Target Information
PTEN은 다양한 암에서 높은 빈도로 돌연변이가 발견된 종양 억제 유전자로, phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase 활성을 갖습니다.
이 단백질은 tensin 유사 도메인과 dual specificity protein tyrosine phosphatases의 촉매 도메인을 포함하고 있으며, 주로 phosphoinositide 기질을 탈인산화합니다.
PTEN은 세포 내 phosphatidylinositol-3,4,5-trisphosphate 수준을 음성 조절하여 AKT/PKB 신호 경로를 억제함으로써 종양 억제 기능을 수행합니다.
또한 PTEN은 성체 신경 발생 과정에서 신경세포의 위치, 수상돌기 발달 및 시냅스 형성을 조절하는 AKT-mTOR 신호 경로의 주요 조절자로 작용합니다.
추가적으로, PTEN은 지방조직에서 인슐린 신호 및 포도당 대사의 음성 조절자로 알려져 있습니다.
핵 내 단일 유비퀴틴화된 형태는 세포자멸을 더 강하게 유도하며, 세포질 내 비유비퀴틴화된 형태는 종양 억제 능력이 상대적으로 낮습니다.
