Target Information
PTEN은 다양한 암에서 변이되는 종양 억제 유전자로, phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase 활성을 가진다. 이 단백질은 tensin 유사 도메인과 이중 특이성 단백질 티로신 포스파타제와 유사한 촉매 도메인을 포함한다. 대부분의 단백질 티로신 포스파타제와 달리, PTEN은 phosphoinositide 기질을 선호적으로 탈인산화하며, AKT/PKB 신호 경로를 음성 조절하여 세포 내 phosphatidylinositol-3,4,5-trisphosphate 수준을 낮추고 종양 억제 기능을 수행한다. 또한 PTEN은 신경 발생 과정에서 성체 신경세포의 통합, 위치 결정, 수상돌기 발달 및 시냅스 형성 조절에 관여하며, 지방 조직에서 인슐린 신호 및 포도당 대사의 음성 조절자로 작용할 수 있다. 핵 내 단일 유비퀴틴화된 형태는 더 높은 세포자멸 유도 능력을 가지며, 세포질 내 비유비퀴틴화 형태는 상대적으로 낮은 종양 억제 능력을 나타낸다.
