Product Specific Information

This antibody reacts with an extracellular antigen of both cell surface-expressed and soluble forms of CD44 antigen (Phagocyte glycoprotein 1), an 80–95 kDa transmembrane glycoprotein (hyaladherin family) present on most cells and tissues such as leukocytes, endothelial cells, and mesenchymal cells. It is negative on platelets and hepatocytes.

Western Blot Protocol Example:

1. Resuspend approximately 50 million cells in 1 mL cold lysis buffer (1% laurylmaltoside in 20 mM Tris/Cl, 100 mM NaCl pH 8.2, 50 mM NaF, including protease inhibitor cocktail).
2. Incubate for 60 minutes on ice.
3. Centrifuge to remove cell debris.
4. Mix lysate with non-reducing SDS-PAGE sample buffer.
5. Perform Western blot under non-reducing conditions for 60 minutes on a vertical incubator.

Target Information

CD44 is a 100 kDa type I transmembrane glycoprotein widely expressed on human leukocytes, white matter of the brain, and some epithelial cells of the intestine and breast. Several isoforms exist, including CD44H, the predominant form in normal tissues. CD44 serves as a receptor for hyaluronic acid (HA) and plays roles in cell-cell interaction, adhesion, and migration. It is involved in various cellular functions such as lymphocyte activation, recirculation, and homing.

CD44 expression may increase in certain carcinomas and is linked to metastatic potential. It is expressed by hematopoietic and non-hematopoietic cells, epithelial tissues, and filopodia in cultured keratinocytes. Bone marrow myeloid and memory T cells express high levels of CD44, and peripheral B and T cells can upregulate CD44 upon stimulation. The CD44 gene undergoes complex alternative splicing, producing functionally distinct isoforms. Diseases associated with CD44 dysfunction include superficial keratitis and lichen sclerosus. CD44 may also be related to tumor metastasis formation.
