Product Specific Information

Description:
The W6/32 monoclonal antibody reacts with human MHC class I antigens (HLA-A, B, C). These antigens, associated with beta 2-microglobulin, are expressed on all nucleated human cells and play a key role in immune response and tumor surveillance. The W6/32 clone recognizes a non-polymorphic epitope shared among HLA-A, B, and C, and immunoprecipitates both 43 kDa and 11–12 kDa chains. Cross-reactivity is observed in baboon, rhesus, and cynomolgus monkeys.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-diluted and tested for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (1.0 µg) per test in 100 µL final volume. Optimal cell number: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended detection filter: 610/20 bandpass.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions. Protect from light due to photo-induced oxidation sensitivity.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Excitation/Emission:
405 nm / 600 nm (Violet Laser)

HLA-class I MHC antigens are membrane glycoproteins expressed on nucleated cells, associated with beta2 microglobulin. They present foreign peptides for recognition by cytotoxic T cells, essential for antiviral and antitumor immune defense.
HLA and MHC antibodies are key tools in Immunopeptidomics, supporting identification and characterization of neoantigens via LC-MS/MS.
