CD26 (dipeptidyl peptidase IV, DPP IV, adenosine deaminase binding protein) is a homodimeric atypical serine protease belonging to the prolyl oligopeptidase family. It is expressed on lymphocytes and upregulated during T-cell activation, also found on activated B cells, NK cells, and epithelial cells. CD26 participates in T-cell activation, cell adhesion to extracellular matrix (e.g., fibronectin, collagen), and HIV infection. It is identical to adenosine deaminase complexing protein-2 and functions as a serine exopeptidase that cleaves X-proline dipeptides from the N-terminus of polypeptides. Alterations in CD26 activity are linked to malignant transformation, with increased enzymatic activity correlating with tumor grade. CD26 expression changes have been observed in monocytes and CD4+ T cells during rheumatoid arthritis and systemic lupus erythematosus.
