Product Specific Information
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, mature T helper cells, and monocytes. CD4 serves as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 recognize distinct epitopes.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on human peripheral blood cells. Recommended use: 5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample before combining with cells.
- For single-color controls, use 5 µL per 100 µL of cell sample.
NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information
CD4 antigen is a co-receptor for HIV and recognizes MHC class II molecules. It is expressed primarily on T helper cells, but also on monocytes, macrophages, dendritic cells, and various tissues such as thymus, lymph nodes, tonsils, spleen, and specific regions of the brain and gut.
CD4 plays a role in early T-cell activation via association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in infectious and immune-mediated CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
