Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, mature T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV and SK3 blocks HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:

• Flow cytometric analysis

Applications Tested:

• Pre-titrated and tested on normal human peripheral blood cells
• Recommended use: 5 µL (0.1 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) should be added to antibody cocktails prior to combining with cells (5 µL per sample).
• For single-color controls: add 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton technology with engineered nucleic acid dye structures, designed to minimize spillover and spread.

Target Information

CD4 is a co-receptor for MHC class II molecules and HIV. It is primarily expressed on T helper cells and also found on monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and various non-lymphoid tissues. CD4 aids early T-cell activation via TCR complex and Lck kinase interaction and may play a role in neuronal damage during immune-mediated CNS diseases.
