Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa surface receptor expressed on most thymocytes, T helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV, and SK3 can block HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 recognize distinct epitopes.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.
This antibody is pre-titrated and validated for flow cytometric analysis of human peripheral blood cells. Recommended usage: 5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology, engineered for optimized fluorescence with reduced spillover and spread effects.

Target Information

CD4 is a co-receptor for MHC class II recognition and HIV entry. It is expressed on T helper cells, monocytes, macrophages, and dendritic cells, and found in thymus, lymph nodes, tonsils, spleen, and other tissues. CD4 plays a key role in T-cell activation via association with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
