Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV, and SK3 can block HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 recognize distinct epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for normal human peripheral blood cells at 5 µL (0.1 µg)/test.
A test is defined as staining of cells in 100 µL final volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails prior to cell addition.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology with engineered nucleic acid dye structures, designed for optimized fluorescence with minimal spillover and spectral spread.

Target Information

CD4 is a co-receptor for MHC class II recognition and HIV entry, primarily expressed on T helper cells, monocytes, macrophages, and dendritic cells. It plays a role in T-cell activation via association with the TCR complex and Lck kinase. CD4 expression is found in lymphoid tissues (thymus, lymph nodes, tonsils, spleen) and certain non-lymphoid tissues (brain, gut). Multiple alternatively spliced transcripts exist (RefSeq, July 2017).
