This antibody detects endogenous levels of p53 only when phosphorylated at Serine 20.


Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation leads to transcription of genes such as p21waf1, regulating G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 and Ser20 is critical for p53 activation and stabilization. p53 functions as a key anticancer factor by blocking proliferation of damaged cells. Phosphorylation at Ser392 is associated with tumor formation. Additionally, p53 is implicated in aging, oxidative stress, and neural function, with increased levels linked to learning and memory deficits.

In normal cells, p53 expression is low, but it is upregulated in transformed cell lines, contributing to malignancy. Mutations in TP53 disrupt DNA binding and tumor suppressor function, commonly found in various human cancers. Germline mutations in TP53 are also linked to Li-Fraumeni syndrome.
