Product Specific Information

Antibody detects endogenous levels of p53 only when phosphorylated at Serine 46.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 up-regulates stress-responsive genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 comprises an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation of p53 at various sites—including Ser15 and Ser392—affects its activation, stabilization, and association with tumor formation. p53 plays crucial roles in preventing proliferation of damaged cells, acting as an anticancer factor, and is also linked to aging and oxidative stress. Mutations in TP53 are common in human cancers and result in loss of tumor suppressor activity. Germline mutations can lead to hereditary cancer syndromes such as Li-Fraumeni syndrome.
