Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. CD3e, together with CD3γ and CD3δ, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed on thymocytes and all mature T cells. Crosslinking via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.

NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays with NovaFluor dyes.

Each NovaFluor conjugate or kit is shipped with CellBlox Blocking Buffer.
Use 5 µL per sample when staining with NovaFluor conjugates or antibody cocktails.
For single-color controls, add 5 µL CellBlox Blocking Buffer per 100 µL of cell sample (10³–10⁸ cells).

Target Information

The CD3 complex is essential for transmitting antigen-recognition signals from the TCR into the cytoplasm and regulating TCR surface expression. It includes CD3γ, CD3δ, CD3ε, and CD3ζ subunits, encoded on chromosome 11. These subunits contain cytoplasmic tails with tyrosine-based motifs that mediate association with signaling molecules.

CD3 is expressed on:

• 68–82% of normal peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

It is not expressed on B cells or NK cells.
Altered CD3 expression may be observed in certain autoimmune diseases.
Defects in the CD3 gene are associated with CD3 immunodeficiency.
