Product Specific Information
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The CD3 epsilon chain, along with gamma and delta subunits, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.1 µg)/test in 100 µL final volume. Cell count range: 10⁵–10⁸ cells/test.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample to antibody cocktails prior to combining with cells.
- For single-color controls: use 5 µL of CellBlox Blocking Buffer per 100 µL of cell sample.
NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information
CD3 complex transduces antigen-recognition signals into T cell cytoplasm and regulates TCR surface expression. The CD3 subunits (gamma, delta, epsilon, zeta) are members of the immunoglobulin superfamily encoded on chromosome 11. CD3 is expressed on most T lymphocytes and thymocytes, but not on B or NK cells. Mutations in CD3 genes can lead to immunodeficiency.
