Product Specific Information

Description:
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. CD3e, together with gamma and delta chains, is essential for proper TCR assembly, trafficking, and surface expression. CD3 is expressed by thymocytes and mature T cells. Crosslinking via immobilized UCHT1 induces intracellular signaling leading to activation and proliferation.

Applications Reported:
Flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.6 µg) per test (100 µL total volume, 10⁵–10⁸ cells/test).

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains instead.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) enhances staining performance. Add 5 µL per sample to antibody cocktails or single-color controls.

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures minimizing spectral spillover and spread.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded on human chromosome 11. These subunits associate with signal transduction molecules via cytoplasmic motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. CD3 gene defects are linked to immunodeficiency.
