Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed on thymocytes and mature T cells.

Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Use 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Improved formulation over CellBlox.
• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are immunoglobulin superfamily members encoded on chromosome 11.

CD3 is expressed on:

• 68–82% of normal peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

Not expressed on B or NK cells.
Defects in CD3 genes are associated with CD3 immunodeficiency.
