Description:
The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF-alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4⁺ T cells. TNF-alpha exhibits cytolytic activity against various tumor cells and plays a key role in immune regulation. It forms dimers and trimers and also exists as a 26 kDa membrane-bound form. The MAb11 antibody is a neutralizing antibody.

Applications Reported:
Reported for use in detection of human TNF-alpha by ELISA.

Applications Tested:
Tested as a detection antibody in sandwich ELISA for human TNF-alpha analysis, in combination with purified MAb1 (14-7348) for capture and recombinant human TNF-alpha (14-8329) as standard. Recommended detection concentration: 0.5–2 µg/mL.
Include a standard curve of recombinant TNF-alpha with doubling dilutions from 1000 pg/mL to 8 pg/mL per ELISA plate.

Target Information

TNF-alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF-alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It forms multimers (dimers, trimers, pentamers) and shows a single 17 kDa band under non-reducing SDS-PAGE. Knockout studies in mice suggest neuroprotective functions and tumor necrosis activity. TNF-alpha is involved in immune responses to bacterial, viral, parasitic, and fungal infections, and contributes to tumor necrosis. It acts synergistically with interferon-gamma in cytotoxicity and is implicated in autoimmune diseases, insulin resistance, and cancer.
