The antibody detects a 130 kDa protein corresponding to induced NOS on SDS-PAGE immunoblots of mouse macrophages treated with LPS followed by pervanadate. It also detects a 150 kDa doublet corresponding to higher molecular weight forms of NOS, which are observed using other anti-iNOS antibodies. Additionally, a non-specific 120 kDa band may be detected.

Target Information

iNOS (Inducible Nitric oxide synthase) generates nitric oxide (NO), a gaseous free radical mediating vasorelaxation, neurotransmission, and cytotoxicity. iNOS is expressed in liver and inducible by lipopolysaccharide and cytokines. NOS family members catalyze oxidation of L-arginine to L-citrulline and NO, containing calmodulin, NADPH, FAD, and FMN binding domains. iNOS is found in macrophages, hepatocytes, synoviocytes, and smooth muscle cells. Cytokine stimulation increases iNOS mRNA, protein, and activity levels, leading to sustained NO production. iNOS dysfunction is associated with diseases such as achalasia and impotence.
