Target Information

NANOG (Nanog homeobox) is a divergent homeodomain protein that regulates pluripotency and differentiation of embryonic stem cells.

  • Expressed in pluripotent human and mouse cell lines, absent in differentiated cells.
  • Shares 52% overall amino acid identity with mouse NANOG and 85% identity in the homeodomain.
  • Human NANOG maps to chromosome locus 12p13.31; mouse NANOG maps to locus 6 F2.
  • Plays a key role in the inner cell mass and embryonic stem (ES) cell proliferation and self-renewal.
  • Overexpression promotes S-phase entry and proliferation.
  • Associated diseases: teratocarcinoma and germ cell/embryonal cancer.

NANOG is also involved in the generation of induced pluripotent stem (iPS) cells, which may be useful in regenerative medicine. Expression of NANOG, Sox2, Klf4, and c-Myc can generate iPS cells without viral vectors, improving safety for clinical applications.
