Target Information
This gene generates several transcript variants differing in their first exons. At least three alternatively spliced variants encoding distinct proteins have been reported, two of which encode isoforms functioning as inhibitors of CDK4 kinase.
Another transcript includes an alternate first exon located 20 Kb upstream, encoding an alternate open reading frame (ARF) that produces a structurally unrelated protein.
The ARF product stabilizes tumor suppressor protein p53 by interacting with and sequestering MDM2, an E3 ubiquitin-protein ligase responsible for p53 degradation.
Despite structural and functional differences, both CDK inhibitor isoforms and the ARF product share a role in cell cycle G1 control through regulation of CDK4 and p53.
This gene is frequently mutated or deleted in various tumors and acts as an important tumor suppressor gene.
