Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 upregulates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.
The p53 structure includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerization domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is critical for activation and stabilization of p53. Phosphorylation at Ser392 has been associated with tumor formation. p53 plays roles in tumor suppression, aging, and oxidative stress response. Mutations in the TP53 gene are common in human cancers and can lead to loss of tumor suppressor function. Germline mutations in TP53 are linked to Li-Fraumeni syndrome.
