Product Specific Information
This antibody detects endogenous levels of IRS1 only when phosphorylated at Ser323, which site has been historically referenced as Ser318.
Target Information
IRS-1, a major substrate of the insulin receptor, is phosphorylated in response to stimulation by insulin, insulin-like growth factor 1 (IGF-1), and interleukin 4 (IL-4). It undergoes phosphorylation on serine, threonine, and tyrosine residues in various tissues. Casein kinase II mediates part of the insulin-stimulated serine/threonine phosphorylation of IRS-1 in vivo, with Thr502 and Ser99 identified as major phosphorylation sites. IRS-1 contains three putative binding sites for 14-3-3 proteins (Ser270, Ser374, Ser641), with the Ser270 motif located in the phosphotyrosine-binding domain responsible for interaction with the insulin receptor.
