Target Information

TAK1 (also known as MAP3K7) is a serine/threonine kinase involved in multiple signaling pathways. It modulates TGF-β activation of JNK and p38 through interactions with TRAF6 and TGF-β receptor. TAK1 also participates in Wnt, BMP, activin, thyroid hormone, osmotic stress, endothelin, and ephrine signaling. It promotes wound healing via activation of von Hippel-Lindau tumor suppressor expression and represses PDGF-B and integrins β1/β5. TAK1 is essential for IKK activation in IL-1, IL-6, IL-18, TNF, CD40, TLR, and RIG-I pathways, requiring TAB2/3 and ubiquitination. It is autophosphorylated at Thr178, Thr184, Thr187, and Ser192, and phosphorylates IKK-β. TAK1 also represses human telomerase reverse transcriptase, suggesting a role in cellular lifespan regulation.
