Product Specific Information

This FFA21 monoclonal antibody reacts with mouse interleukin-21 (IL-21), a 17 kDa immunomodulatory cytokine produced mainly by NKT, Th17, and TFH cells.
In TFH cells, IL-21 expression leads to autocrine signaling through IL-21R and STAT3, activating Bcl6 transcription. IL-21 is crucial for TFH effector function and plays a role in B cell differentiation, IgG production, and apoptosis of naive B cells.

In Th17 cells, IL-21 expression with STAT3, IRF4, and RORγt upregulates IL-23R, promoting Th17 maturation and maintenance via IL-23. IL-21 also downregulates Foxp3. Elevated IL-21 levels are observed in colitis models, while IL-21-deficient mice are resistant to colitis induction.

Preliminary data suggest that clone FFA21 recognizes a different epitope than anti-mouse IL-21 clone mhalx21 (Product #51-7213-82).

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Optimization: Compatible with eBioscience IC Fixation and Permeabilization Kit (Product #88-8824-00).
Applications Tested: Tested on Th17-polarized mouse splenocytes restimulated with PMA, ionomycin, and brefeldin A for 5 hours. Recommended ≤0.25 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test (empirically determined).

Target Information

IL-21 is a 17 kDa cytokine primarily produced by NKT, Th17, and TFH cells. It mediates autocrine signaling via IL-21R and STAT3, activating Bcl6 and promoting TFH cell function. IL-21 facilitates T cell-dependent B cell differentiation, IgG production, and apoptosis in naive B cells.
In Th17 cells, IL-21 with STAT3, IRF4, and RORγt induces IL-23R expression, enabling Th17 maturation. IL-21 also suppresses Foxp3 expression. IL-21-deficient mice show resistance to chemically induced colitis, whereas elevated IL-21 levels are observed in colitis models.
