Description:
Monoclonal antibody eBio3A3-N2 reacts with human IL-21, a potent stimulator of NK and cytolytic T cells. IL-21 is a 131 amino acid protein related to IL-2 and IL-15, expressed in activated CD4+ cells. It promotes B-cell proliferation (via CD40 crosslinking), bone marrow progenitor cell proliferation, and NK-cell marker CD56 expression in the presence of IL-15.

Applications Reported:
Reported for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by intracellular staining of restimulated, Th17-polarized normal human peripheral blood cells. Use at 5 µL (0.5 µg) per test. A test is defined as the amount of antibody that stains a cell sample in 100 µL final volume (10⁵–10⁸ cells/test, empirically determined).

Notes:

• Staining for IL-21 shows donor variability; stimulation times may vary and should be optimized.
• Increased staining levels observed on Th17-polarized normal human peripheral blood cells.
• Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.
• Filtration: 0.2 µm post-manufacturing filtered.

Target Information

IL-21 is a 17 kDa immunomodulatory cytokine produced mainly by Natural Killer (NKT), T helper (Th)17, and T follicular helper (TFH) cells.
In TFH cells, IL-21 triggers autocrine signaling via IL-21R and STAT3, leading to Bcl6 activation. Like IFN-γ for Th1 and IL-4 for Th2, IL-21 is critical for TFH cell development and effector function.
IL-21 regulates T cell-dependent B cell differentiation into plasma and memory cells, stimulates IgG production, and induces apoptosis in naive B cells.
In Th17 cells, IL-21 enhances IL-23R expression via STAT3, IRF4, and RORγt, facilitating maturation and maintenance by IL-23. It also downregulates Foxp3.
IL-21 deficient mice are resistant to chemically induced colitis, while elevated IL-21 levels are seen in colitis models.
