- Immunohistochemistry (PFA fixed):
Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD47 (Integrin-Associated Protein, IAP)은 세포 표면에 널리 발현되는 막관통 당단백질로, 여러 인테그린과 상호작용하여 그 기능을 조절합니다.
CD47의 리간드 결합은 G 단백질 신호전달 경로를 조절하며, 세포 부착 시 세포 내 칼슘 농도 증가에 관여합니다. 또한 thrombospondin의 C-말단 결합 도메인의 수용체로 작용하며, 막 수송 및 신호전달에 역할을 합니다.
CD47은 다양한 조직에서 발현되며, Rh 적혈구에서는 발현이 감소합니다. CD47은 대식세포의 억제 수용체인 CD172a(SIRPα)의 리간드로 작용하여 CD47 양성 세포의 식작용을 억제합니다.
또한 CD47–CD172a 시스템은 세포 이동, B 세포 부착, T 세포 활성화, 연골세포의 기계적 신호 반응 조절, 신경 발달에 관여하며, 뇌와 망막의 시냅스가 풍부한 부위에서 특히 많이 존재합니다.
