Target Information
Isoform 1 acts as an extracellular chaperone preventing aggregation of nonnative proteins and stress-induced aggregation of plasma proteins. It inhibits amyloid fibril formation by APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants. Functions without ATP, maintaining proteins for refolding by other chaperones (e.g., HSPA8/HSC70). Secreted isoform protects cells against apoptosis and complement-mediated cytolysis. Intracellular isoforms promote ubiquitination and proteasomal degradation via SCF E3 ubiquitin ligase complexes, modulate NF-κB activity, and regulate apoptosis and cell proliferation.
