Product Specific Information
This TNKUPJ monoclonal antibody recognizes mouse CD163, a 130 kDa surface receptor expressed by subsets of tissue macrophages such as splenic red pulp macrophages, Kupffer cells, intestinal lamina propria macrophages, and a small fraction of peritoneal macrophages. Mouse monocytes do not express CD163, and unlike human CD163, it is not a reliable marker for M2 macrophages.
The antibody detects CD163 on fixed and permeabilized cells, allowing intracellular staining. Aggressive tissue dissociation may reduce surface CD163, in which case intracellular detection is recommended.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes, ≤0.5 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best results.
Fluorochrome Information:
Alexa Fluor 532 is excited with a 532 nm green laser and emits at 561 nm. Not compatible with the 561 nm yellow-green laser. Recommended filter: 560/14 band pass.
Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein, a member of the scavenger receptor cysteine-rich superfamily. It functions as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and aiding iron recycling. CD163 is expressed on human monocytes and macrophages, upregulated by IL-10, IL-6, and dexamethasone, and shed upon stimulation by LPS or PMA. Binding is calcium- and pH-dependent, with higher affinity for multimeric haptoglobin complexes.
