Product Specific Information
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha).
CD127 complexes with CD132 (common gamma chain, γc) to form the IL-7 receptor (IL-7R), a type I glycoprotein (75–80 kDa). It is expressed on immature B cells (early pre-B stage), thymocytes, and most mature T cells, with transient down-regulation upon activation.
IL-7 binding triggers signal transduction via tyrosine kinase pathways, including Jak/STAT, essential for lymphocyte development and homeostatic T-cell proliferation.
IL-7R signaling also regulates TCR locus rearrangement in γδ T cells.
CD127 expression is notably down-regulated on CD4+CD25+ regulatory T cells (Tregs). Cells co-expressing CD4 and CD25 but lacking CD127 are highly enriched for Tregs, defined by Foxp3 expression and in vitro suppressive activity.
Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Target Information
CD127 (Interleukin-7, IL-7) is a glycoprotein regulating lymphopoiesis.
Its receptor (IL7R) is an alpha/gamma chain heterodimer.
- Gamma(c) chain: Activates signal transduction
- Alpha chain: Determines specific signaling events
CD127 promotes proliferation of precursor B cells, thymocytes, T-cell progenitors, and mature CD4+/CD8+ T cells.
Dysfunction is linked to severe combined immunodeficiency (SCID) and T-cell negative/B-cell negative/NK positive SCID.
