Product Specific Information
The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily.
PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease.
PD-1 is expressed predominantly on activated T and B lymphocytes.
Two ligands of PD-1 have been identified: PD-L1 (B7-H1) and PD-L2 (B7-DC). These ligands exhibit overlapping functions and show constitutive expression on some normal tissues and upregulation on activated antigen-presenting cells.
Costaining experiments suggest that eBioJ105 recognizes a different epitope than MIH4 (Product # 11-9969).
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of stimulated normal human peripheral blood cells.
Use at 5 µL (1.0 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Fluorochrome Information:
Super Bright 436 can be excited with the violet laser (405 nm) and emits at 436 nm.
Recommended filter: 450/50 bandpass or equivalent.
Use Super Bright Complete Staining Buffer (Product # SB-4401) when multiple Super Bright dye-conjugated antibodies are used to minimize non-specific polymer interactions.

Target Information
Cell-mediated immune responses are initiated by T lymphocytes that are stimulated by peptides bound to MHC molecules on antigen-presenting cells (APC).
Activated T cells express inhibitory receptors such as PD-1 (PDCD-1), which modulate immune activation by binding to PD-L1 and PD-L2.
This interaction inhibits immune responses in the absence of danger signals such as LPS.
PD-1 knockout mice exhibit hyperactivated immune systems and autoimmune diseases.
Despite its predicted molecular weight, PD-1 often migrates at higher molecular weight in SDS-PAGE.
