Product Specific Information

This F38-2E2 monoclonal antibody reacts with human CD366 (TIM3 or HAVCR2). TIM3 is expressed on activated CD4+ T cell subsets (Th1, Th17, Treg), CD8+ T cells, monocytes, dendritic cells, and mast cells. It exists as membrane-bound and soluble forms. Galectin-9 is identified as its ligand and plays a role in negative regulation of CD4+ T cells, cytokine secretion, and immune tolerance. TIM3 is implicated in autoimmune diseases such as multiple sclerosis and in HIV infection.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry of stimulated human peripheral blood cells at 5 µL (0.25 µg)/test.
A test is defined as the amount of antibody staining a cell sample in 100 µL volume, with cell counts ranging from 10⁵–10⁸ cells/test.

Fluorochrome Properties:
Super Bright 436 is excited with a violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

TIM3 (HAVCR2, T-cell immunoglobulin and mucin domain-containing protein 3) is a 281 amino acid, type-1 Th1-specific surface glycoprotein expressed on terminally differentiated CD4+ Th1 and CD8+ Tc1 cells. It includes an IgV-like domain, mucin-like extracellular domain, and cytoplasmic tyrosine phosphorylation motif. TIM3 regulates macrophage activation and autoimmune responses, down-regulates Th1-mediated immunity, and contributes to immune tolerance. It is associated with experimental autoimmune encephalomyelitis (EAE) and other Th1-dependent diseases.
