Product Specific Information

The monoclonal antibody 29A1.4 recognizes mouse NKp46 (CD335), a member of the natural cytotoxicity receptor (NCR) family.
CD335 is a glycoprotein with two Ig-like domains and a short cytoplasmic tail, uniquely expressed on NK cells (including immature NK cells, DX5⁻ CD3⁻), allowing discrimination between NKT and NK cells (NKp46⁺, CD3⁻).

Staining with 29A1.4 is not strain-specific, observed in C57Bl/6, SJL, CBA/CA, and BALB/C strains. NKp46 plays a role in NK cell-mediated lysis of tumor and pathogen-infected cells.

• Activation: 29A1.4 activates NK cells in vitro.
• Depletion: Does not deplete NK cells in vivo.
• Applications Reported: Flow cytometric analysis.
• Applications Tested: Tested by flow cytometric analysis of mouse splenocytes (≤1 µg/test).

A test is defined as the amount of antibody staining a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test. Optimal titration recommended.

Fluorochrome Information:
Super Bright 436 is excited with the violet laser (405 nm) and emits at 436 nm. Use a 450/50 bandpass filter or equivalent. Ensure instrument compatibility.

Staining Panels:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product # 00-8222)
• 1-step Fix/Lyse Solution (Product # 00-5333)
  for up to 3 days at 4°C in the dark with minimal brightness impact.
