Product Specific Information
The 4S.B3 monoclonal antibody reacts with interferon-gamma (IFNγ). Human IFNγ is a 17 kDa factor produced by activated T and NK cells, exhibiting antiviral and antiparasitic activity. In synergy with TNFα, it inhibits proliferation of normal and transformed cells and modulates immune responses across various cell types expressing IFNγ receptors. Glycosylation does not affect IFNγ biological activity.
Applications Reported: Intracellular flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of activated human cells at ≤0.125 µg/test (100 µL final volume). Recommended to titrate for optimal performance.
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
IFNγ (Interferon gamma, Type II interferon) is a macrophage activation factor produced mainly by T lymphocytes and NK cells in response to antigens and cytokines. It is a dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits. Human IFNγ is species-specific and does not cross-react with mouse IFNγ.
Functions include:
- Antiviral and tumor antiproliferative activity
- Induction of MHC class I and II
- Macrophage activation
- Enhanced immunoglobulin secretion by B lymphocytes
Activation occurs via IFNγ receptor I and II, triggering the JAK-STAT pathway. Human IFNγ shares ~40% sequence homology with mouse IFNγ. Upregulated by IL2, FGF basic, EGF; downregulated by vitamin D3 or DMN. Mutations are associated with aplastic anemia.
