Product Specific Information
This antibody recognizes an epitope near the C-terminal region (404–441 aa) of human Tau.
It reacts with all seven isoforms of human Tau (both binding-competent and binding-incompetent), A68 (an abnormal phosphorylated derivative of Tau), bovine Tau, and human NeuroFibrillary Tangles (NFTs).
Reactivity is independent of the phosphorylation state of Tau. Some cross-reactivity with MAP2 is also observed.
This antibody is suitable for immunohistochemical staining of Bouin’s-fixed or alcohol-fixed, paraffin-embedded, or frozen tissue sections.
Target Information
Tau is a neuronal microtubule-associated protein predominantly found on axons. Its primary function is to promote tubulin polymerization and stabilize microtubules.
- The C-terminus binds axonal microtubules, while the N-terminus interacts with neural plasma membrane components, acting as a linker between both structures.
- Axonal polarity is determined by Tau/MAPT localization within the neuronal cell body.
- Short isoforms provide cytoskeletal plasticity, while longer isoforms contribute to stabilization.
- In its hyper-phosphorylated form, Tau constitutes paired helical filaments (PHF), the main component of neurofibrillary lesions in Alzheimer’s disease (AD).
- Hyper-phosphorylation disrupts Tau’s microtubule-binding function, leading to cytoskeletal destabilization and neuronal degeneration.
- Multiple kinases phosphorylate Tau, including GSK-3β, PKA, cdk5, and casein kinase II.
- Hyper-phosphorylated Tau is found in neurofibrillary lesions associated with Alzheimer’s disease, Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
