Target Information

This gene encodes a member of the tumor necrosis factor (TNF) cytokine family, which is a ligand for osteoprotegerin and functions as a key factor for osteoclast differentiation and activation. It acts as a dendritic cell survival factor and regulates T cell-dependent immune responses. Activation of T cells induces expression of this gene, leading to increased osteoclastogenesis and bone loss. RANKL activates antiapoptotic kinase AKT/PKB through a signaling complex involving SRC kinase and TRAF6, suggesting a role in cell apoptosis regulation. Gene disruption in mice leads to severe osteopetrosis, lack of osteoclasts, and defects in T/B lymphocyte differentiation and mammary gland development. Two alternatively spliced transcript variants have been identified.
