Product Specific Information
This monoclonal antibody recognizes the cleaved C-terminus of ROCK-1 and does not recognize the full-length protein. Since ROCK-1 is cleaved by caspase-3 during apoptosis, this antibody can be used for the detection of apoptosis.
A suggested positive control is Jurkat cells treated with camptothecin or anti-Fas antibody to induce apoptosis.
Target Information
Rho-associated coiled coil-containing protein kinase (ROCK)-1 is a substrate of caspase-3 and an effector of the small GTPase Rho. It contributes to myosin light chain (MLC) activation via direct phosphorylation and inhibition of MLC phosphatase.
During apoptosis, ROCK-1 is cleaved by caspase-3 at a conserved DETD1113/G sequence, removing its inhibitory domain and resulting in constitutive kinase activity. This cleavage leads to phosphorylation of MLC and membrane blebbing, key events in the execution phase of apoptosis.
