Product Specific Information
The A20 monoclonal antibody reacts with mouse CD45 (leukocyte common antigen, LCA) in CD45.1-expressing strains such as SJL/J, DA, STS/A, and RIII. CD45.1 is expressed on all leukocytes in these strains.
- Applications Reported: Flow cytometric analysis
- Applications Tested: Tested on mouse splenocytes at ≤ 0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Titration recommended for optimal performance.
- Fluorochrome: Brilliant Violet™ 605 (BV605) emits at 605 nm; use with violet (405 nm) laser.
- Buffer Recommendation: When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.
- Light Sensitivity: Protect from light to prevent photo-induced oxidation.
- Fixation: Compatible with IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54); store up to 3 days at 4°C in dark. Not compatible with methanol-based fixation.

Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It regulates Src-family kinases, plays a role in lymphocyte development and antigen signaling, and modulates integrin-mediated pathways.
Alternative splicing produces multiple isoforms differing in extracellular domains, influencing cell-type-specific expression and membrane localization. CD45RA is one such isoform with restricted expression among lymphoid subtypes.
