Target Information
NUMBL plays a role in neurogenesis. It is required throughout embryonic neurogenesis to maintain neural progenitor cells (radial glial cells, RGCs) by allowing their daughter cells to choose progenitor over neuronal fate. It is not required for proliferation of neural progenitor cells before embryonic neurogenesis onset. Postnatally, it regulates SVZ neuroblast survival and ependymal wall integrity. NUMBL acts as a negative regulator of NF-kappa-B signaling pathway by preventing MAP3K7IP2 interaction with polyubiquitin chains of TRAF6 and RIPK1, and by promoting Lys-48-linked polyubiquitination and degradation of TRAF6 in cortical neurons.
