Target Information

Diacylglycerol (DAG) acts as an intracellular activator of protein kinase C (PKC) and guanine nucleotide exchange factors, influencing various cell signaling cascades. Intracellular DAG levels are regulated by diacylglycerol kinases (DGKs), which convert DAG to phosphatidic acid.
Human DGK isoforms are classified as follows:

• Type I: DGK-α (80 kDa), DGK-β (90 kDa), DGK-γ (90 kDa) — contain calcium-binding EF-hand motifs
• Type II: DGK-δ (130 kDa), DGK-ι (130 kDa) — contain N-terminal pleckstrin homology (PH) domains
• Type III: DGK-ε (64 kDa) — lacks identifiable regulatory domains
• Type IV: DGK-Ω (104 kDa), DGK-ι (130 kDa) — possess C-terminal ankyrin repeats
• Type V: DGK-θ (110 kDa) — contains 3 cysteine-rich domains and a PH domain
