Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events such as the establishment and maintenance of pregnancy. PR exists as two major forms:
- Form A (94 kDa)
- Form B (120 kDa)
PR-B is the transcriptionally active form responsible for activating genes related to endometrial maintenance, pregnancy, and inhibition of ovulation. PR-A lacks 165 amino acids at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, the receptor undergoes a conformational change, dimerizes, and binds to progesterone response elements (PRE), leading to transcriptional activation. Mutations in the PGR gene can cause reproductive abnormalities.
