Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH).
This tag is commonly expressed at either the N- or C-terminal regions of recombinant proteins to enable isolation or purification by immobilized metal affinity chromatography (IMAC).
Epitope tagging allows detection of proteins lacking specific antibodies by fusing a known epitope to the protein of interest.
His-tagged proteins can be purified using metal chelate chromatography due to the strong and selective binding of histidine clusters to Ni²⁺ ions on Ni²⁺-NTA adsorbents.
His tag-specific antibodies facilitate detection or co-immunoprecipitation of tagged proteins.
