Product Specific Information

This KIKSI0803 monoclonal antibody recognizes STAT1 phosphorylated on tyrosine 701. STAT proteins are activated by cytokine receptor binding and phosphorylation by JAK family kinases, leading to nuclear translocation and DNA binding for gene regulation. Phosphorylation of STAT1 on Tyr701 is essential for dimer formation and transcriptional activity.

Specificity was confirmed by ELISA, flow cytometry, and western blot.
Reported applications include intracellular staining followed by flow cytometric analysis.
Tested for use at 5 µL (0.125 µg) per test, suitable for 10⁵–10⁸ cells/test.

Recommended Protocol:
Use Protocol C: Two-step (Fixation/Methanol).
Protocols A and B are not compatible.

Fluorochrome Information:
eFluor™ 450 is an alternative to Pacific Blue™, excited by Violet laser (405 nm) and emits at 445 nm. Ensure instrument compatibility.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

STAT1 is a transcription factor activated by interferons (α, β, γ), EGF, PDGF, IL6, and other cytokines. It regulates genes involved in cell growth, apoptosis, immune response, and lipid metabolism. Located on chromosome 2, STAT1 activation involves phosphorylation by JAK kinases, leading to nuclear translocation and transcriptional regulation.
Among STAT family members, STAT1, STAT3, STAT5a, and STAT5b show broad activation profiles. Two alternatively spliced isoforms of STAT1 have been described.
