Target Information
GAD-65와 GAD-67은 group II decarboxylase 계열 단백질로, L-glutamic acid로부터 GABA(gamma-aminobutyric acid)를 생성하는 속도 제한 단계(rate-limiting step)를 촉매합니다.
두 단백질 모두 뇌에서 발현되지만, GAD-65는 신경 말단의 시냅스 소포막에 존재하며, GAD-67은 세포 전체에 분포합니다.
GAD-67은 기본적인 GABA 합성에 관여하며, 신경전달 시 GABA 요구량이 증가할 경우 GAD-65가 일시적으로 활성화되어 GABA 생산을 돕습니다.
GAD-65 결손은 GABA 신경전달에 장애를 일으키지만, GAD-67 결손은 치명적입니다.
대체 스플라이싱으로 인해 GAD-67에는 주요 형태(GAD-67)와 부형태(GAD-25) 두 가지 isoform이 존재합니다.
