Product Specific Information
Description:
The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Binding of 17A2 to CD3 initiates intracellular biochemical pathways resulting in cellular activation and proliferation.
Applications Reported:
The 17A2 antibody is reported for use in flow cytometric analysis and in vitro activation of mouse T cells. For functional assays, use the Functional Grade purified 17A2 (Product #16-0032).
Applications Tested:
Tested by flow cytometric analysis of mouse thymocytes and splenocytes. Recommended ≤0.25 µg per test (100 µL final volume). Optimal antibody concentration should be titrated for each assay.
Purity: >90% (SDS-PAGE)
Aggregation: <10% (HPLC)
Filtration: 0.2 µm post-manufacturing filtered
Target Information
The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR complex expression. CD3 subunits (gamma, delta, epsilon, zeta) are structurally related members of the immunoglobulin superfamily encoded on human chromosome 11. They associate with cytoplasmic signal transduction molecules via a double tyrosine-based motif. CD3 plays a role in TCR-induced growth arrest, survival, and proliferation. CD3 antigen is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
