Target Information

HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under reduced oxygen tension. HIF-1 is a heterodimer consisting of alpha and beta subunits, both belonging to the basic-helix-loop-helix PAS family of transcription factors.

Under hypoxic conditions, HIF-1 activates transcription of over 40 genes involved in oxygen delivery and metabolic adaptation, including erythropoietin, glucose transporters, glycolytic enzymes, VEGF, and HILPDA.

HIF1-alpha regulates hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. It interacts with p53 and MDM2 under hypoxia, influencing ubiquitination and degradation. HIF1A also induces NOX4 to maintain ROS levels and promote proliferation after hypoxia.

In humans, HIF1A is located on chromosome 14q. Its C-terminal binds p300, forming complexes that induce hypoxia-responsive genes such as VEGF. Hypoxia plays a critical role in diseases including ischemia, cancer, pulmonary hypertension, congenital heart disease, and COPD.
