Target Information
HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under reduced oxygen tension. HIF-1 is a heterodimer consisting of alpha and beta subunits, both belonging to the basic-helix-loop-helix PAS family of transcription factors. It regulates transcriptional responses to hypoxia, activating over 40 genes involved in oxygen delivery and metabolic adaptation.
HIF1-alpha is implicated in hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. Under hypoxic conditions, p53 interacts with HIF1-alpha, promoting MDM2-mediated ubiquitination and degradation. Induction of NOX4 by HIF1-alpha helps maintain ROS levels after hypoxia. In humans, HIF1A is located on chromosome 14q, and its C-terminal binds p300, forming complexes that induce hypoxia-responsive genes such as VEGF.
Hypoxia plays a major role in diseases including myocardial and cerebral ischemia, cancer, pulmonary hypertension, congenital heart disease, and chronic obstructive pulmonary disease.
