Product Specific Information
The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Binding of 17A2 to CD3 initiates intracellular biochemical pathways resulting in cellular activation and proliferation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Use ≤0.5 µg/test (100 µL final volume). Cell number can range from 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.
Fluorochrome Information:
eFluor® 506 is excited with the violet laser line (405 nm) and emits at 506 nm. Recommended filter: 510/20 bandpass or equivalent.
Excitation: 405 nm; Emission: 506 nm; Laser: Violet.
Post-manufacturing filtration: 0.2 µm.
Target Information
The CD3 subunit complex is crucial in transducing antigen-recognition signals into the cytoplasm of T cells and regulating TCR surface expression. CD3 gamma, delta, epsilon, and zeta subunits are structurally related members of the immunoglobulin superfamily encoded on chromosome 11. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
