Product Specific Information

Description:
The 104 monoclonal antibody reacts with the mouse CD45 molecule, the leukocyte common antigen (LCA) in CD45.2-expressing mouse strains. Strains expressing CD45.2 include C57BL/6, BALB/c, C58, DBA/1, DBA/2, C3H/He, CBA, 129, A, and AKR. CD45.2 is expressed by all leukocytes in these strains.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometry using mouse splenocytes. Recommended ≤ 1 µg/test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test. Optimize concentration for each assay.

Fluorochrome Information:
APC-eFluor 780 emits at 780 nm, excited by the red laser (633 nm). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD45.2 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It plays a crucial role in lymphocyte development and antigen signaling by regulating Src-family kinases.
CD45.2 exists in multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. These isoforms vary in membrane localization and expression based on cell type and physiological state.
Beyond immunoreceptor signaling, CD45.2 contributes to cell survival by modulating integrin-mediated signaling and is involved in DNA fragmentation during apoptosis. CD45RA is an isoform with restricted expression among lymphoid subtypes.
