Description:
The dG9 antibody clone reacts with human perforin (pore-forming protein, pfp). Perforin is one of the cytolytic mediators present in the cytoplasmic granules of cytotoxic T lymphocytes (CTL) and natural killer (NK) cells. It plays a key role in immune responses against tumors and viral infections.

Applications Reported:
Reported for use in intracellular staining followed by flow cytometric analysis, immunoprecipitation, and immunohistochemical staining. Fluorochrome-conjugated dG9 (delta G9) is recommended for intracellular flow cytometry.

Applications Tested:
Tested by intracellular staining and flow cytometric analysis of normal human peripheral blood cells. Recommended usage is ≤0.25 µg per test (100 µL final volume). Optimal titration is advised for specific assays.

Target Information

Perforin is a major cytolytic protein stored in cytoplasmic granules of cytotoxic lymphocytes. It mediates immune defense against tumors and viral infections. The human perforin protein consists of 555 amino acids with a 21 amino acid signal peptide and has a molecular weight of 70–75 kDa. It forms transmembrane pores similar to complement component C9. Expression is restricted to killer cell lines and absent in helper T lymphocytes or other tumor cells. Defects in the perforin gene cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare autosomal recessive disorder. Alternative splicing results in multiple transcript variants.
