Product Specific Information

The 12G5 monoclonal antibody reacts with human CD184 (CXCR4), also known as Fusin, LESTR, or HUMSTR. CD184 is a G-protein-coupled chemokine receptor with seven membrane-spanning domains and functions as a co-receptor for X4 HIV-1 entry into CD4+ cells. It is predominantly expressed on naive T cells and upregulated by PHA and IL-2 stimulation.

The 12G5 antibody partially inhibits chemotaxis and calcium influx induced by SDF-1, blocks CD4-independent HIV-2 infection, and inhibits CD4-dependent infection by certain T-tropic HIV-1 isolates.

Applications Reported:

• Flow cytometric analysis
• Functional blocking of CXCR4 (use Functional Grade purified 12G5, Product #16-9999-81, for functional assays)

• Tested by flow cytometric analysis of normal human peripheral blood cells
• Recommended ≤0.5 µg/test (100 µL final volume)

Target Information

CXCR4 (Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains. It acts as a co-receptor for X4 HIV-1 and HIV-2 entry into CD4+ cells via gp120-mediated fusion. CXCR4 is the receptor for CXCL12/SDF-1, mediating calcium influx and MAPK activation. It also binds extracellular ubiquitin, modulating intracellular signaling.

CXCR4 plays roles in:

• Hematopoiesis and cardiac development
• Gastrointestinal vascularization and endothelial remodeling
• Neuronal survival and cerebellar development
• LPS-induced inflammatory response and TNF secretion by monocytes

Antibodies to CXCR4 block HIV-1 and HIV-2 fusion and infection. The amino-terminal domain and second extracellular loop serve as HIV binding sites.
