The Human Thrombomodulin Simplex ProcartaPlex Kit measures Thrombomodulin protein and is designed to be combinable with other Simplex kits, allowing creation of a custom multiplex panel utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed per assay plate regardless of plex size.

ProcartaPlex immunoassays are based on sandwich ELISA principles, using two highly specific antibodies binding to different epitopes of one protein to quantitate all targets simultaneously with a Luminex instrument. Only 25 μL of plasma or serum, or 50 μL of cell culture supernatant is required, and results are obtained within four hours.

Technology Overview

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single sample.
Luminex beads are internally dyed with precise proportions of red and infrared fluorophores to create spectrally unique signatures identifiable by Luminex detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).

Each bead is conjugated with antibodies specific for a single target protein. Bound proteins are detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The fluorescence intensity correlates with protein quantity.

Unlike traditional ELISA, the capture antibody in ProcartaPlex assays is bead-conjugated and free-floating in solution. Instruments such as Luminex 200 use dual lasers—one for bead identification and one for quantifying RPE fluorescence—enabling multiplex profiling with minimal sample volume.

ProcartaPlex multiplex panels are available for six species (human, mouse, rat, nonhuman primate, porcine, canine).
